In most cases, mint stamps, those that have not been used, are worth more. In a few cases, used stamps are more valuable. Modern used stamps are typically worth very little, but there are exceptions and the most valuable stamps in the world are used.

It depends very much on the specific stamp in question. For the most part, unused stamps, referred to as Mint stamps, are more valuable then used ones. In some cases a used stamp on a cover may have more value. There are several reasons for this. It may be a First Day of Issue, with a concellation that indicates that the stamp was used on the first day it was available. Noteworthy dates and place may be of much more value then the stamp, for instance a cover postmarked December 7th, 1942 in Pearl Harbor. Some foreign stamps that are issued by countries that are just trying to make money by making and selling stamps are worth more when they are used for actually mailing a letter. And some US stamps, such as the Military Postage stamps used by the Recruiting command are worth more on cover than mint.
