In the first Sister Act, the characters are mostly all from a Convent which means that those women are all Sisters of God. Whoopi Goldberg is not from the Convent but is a lounge singer who was the girlfriend of a mob guy in Las Vegas. She is hiding in the Convent after she witnessed her boyfriend murder a man. So the cops are hiding her until the trial where she will testify. To blend in, Whoopi has to become a nun (Sister Mary Clarence) and the only ones who knows she is not are the Mother Superior and the cop assigned to her. The movie is about her blending in with the convent and helping them in the choir while aggravating the Mother Superior. It's really a funny movie.
