it is only important for special moves. it means push all three punch buttons. when not doing a special move it just registers as Lpunch.

Hadoken input is always quarter-circle forward followed by any punch button.
If you mean Shoryuken then just do the movement you need to do for a Hadoken twice and press the hit button. (down, forward, down, forward, punch)
Press and hold 'back' on your directional pad/analog stick for about a second and a half and then press forward and a punch button.

Akuma does not have a 100 hit combo in Street Fighter IV called "The Dark Hado". He does have a move that is called the Shin Shun Goku Satsu though, which does 27 hits. You can do that by getting a half-full to full revenge meter then pressing in sequence: weak punch, weak punch, back, weak kick, strong punch. He also has a super version of it called the Shun Goku Satsu too that does 15 hits. The way you do that, is you have to first get a full super bar then you must press: weak punch, weak punch, forward, weak kick, strong punch. For both of these moves, you have to enter the sequence quite fast. I hope that helps.
