Jerusalem (or Al-Quds) - (Both Mecca & Madina preceeding Jerusalem in Importance)

The last Prophet of Islam Hazrat Muhammad SAW was forced to migrate to Madina Munawara from Mecca Mukarramah in 622. Most of the Muslims also migrated from Mecca.
Mecca where the Ka'abah is located is in Saudi Arabia. The last Prophet of Islam Hazrat Muhammad (SAW) was born in Mecca. Madina Munawara is also in Saudi Arabia where the holy Prophet (SAW) is buried. For these reasons Arabia is the most important country for the Muslims. They also travel to Arabia to perform Hajj.

If you see Islam from a historical perspective, yes. Mohammed founded Islam in Mecca. However, according to Islamic theology, Islam has existed since the Creation of the Universe and, therefore, did not start in Mecca.
