From the small amount of info I could find, I think it has to do with a childhood injury. What that injury was and when it happened I have no idea.

from what i know and read his partner was killed or died suddenly and Johnny Depp went into a mental break down and couldn't handle it which then caused him to develop Post-Traumatic Stress Disorder.
