Mespirt is always on the move but if you check your map in your pokewatch thinging on the bottom of the screen there will be a flashing dot and that will be Mespirt or another legendary pokemon!

When Mesprit runs away in Pokemon Diamond or Pearl, it does not settle in one particular place. By using a certain Poke App (obtainable from the Poketch Company building 1F), you can track Mesprit's movements to eventualy encounter it again. Keep in mind that Mesprit will try to run away from you and will change places everytime you enter a new Route or City. Do not fly to where Mesprit is, as it will flee to the other side of Sinnoh. Learn Mesprit's patterns of movement and soon you will be able to encounter it. (Also note that Mesprit runs away during its turn to attack).
Visit Mesprit at Lake Verity (near the starting town) at the cave in the centre. It will fly off, you should be able to track it with your Pokegear.

You go to them, save, and turn off your game until they are shiny. Unless you want to turn off your ds about 24,000 times, I would recommend just living with having unshiny Uxie, Azelf, and Mesprit. By the way, Mesprit is so hard to catch anyway that it would be incredibly rare to find a shiny one.

After going to the little island in lake Verity, Mesprit will fly off. Use the Map app on the Poketch. There will be a little face. That is where Mesprit is. If you fing him on the grass in that route, be careful not to knock him out. He will run off as his move. Make sure to use a faster Pokemon.
