First they will check your vision, if you need correction based on that test they do a refraction (the machine with all the lenses where they ask you "better 1, or better 2?". After they get your glasses rx, if needed, they will check your intraocular pressure using either a tonopen, applanation or non-menthol contact applanation (the air puff), but they will put a numbing drop in your eye so you won't feel anything. Next they dilate your eye (make your pupil big) and then check the back of your eye (retina, optic nerve, macula, blood vessels). A lot depends on age and health, but those are the basic things they look for in an regular eye exam.
