Fallout New Vegas is based in the years after the events of Fallout 1 & 2. Particularly, the NCR has prospered into a huge government will pre-war values of democracy. They needed more resources so they traveled East so the Colorado River and seized the Hoover Dam. Other factions such as the Gun Runners and the Followers of the Apocalypse have also grown over the years.
Essentially, FNV is live the after years of Fallout 2, but in a different setting with an independent storyline.

Nothing in New Vegas spoils Fallout 3. They take place in 2 completely different corners of the country. And New Vegas can not spoil anything for Fallout 3, as New Vegas takes place before the events of Fallout 3. Hope this helps! Also Fallout 3 is not a sequal. But lets pray for a Fallout 4
