Synthetic urine, also known as Agent X is detectable in a DOT drug screening. It will show up in a drug testing lab.
Valium and xanax are benzodiazepines and are not tested for on a DOT drug screen at all. In fact, it is illegal to test for these substances on a federal DOT test. The DOT drug screen looks for marijuana, opiates, PCP, amphetamines and cocaine. Considering that most drivers who do drugs are doing so to stay awake, why would any trucker do downers?
To get a CDL, you'll have to get a DOT physical done, which will include a drug screen.
The DOT does not test for anabolic steroids. A pre-employment or random drug test by the DOT only test for narcotics.
