The bank staff can know the bank which the cheque came from, and, if the cheque is genuine, they can know who is to receive the money. They may also know who has written the cheque by reference to the signature. Bank staff try to learn the signatures.

The account number on the front of the check tells the bank whose account to deduct the amount of the check from.

If you mean what does it do for the bank when you write your account number underneath your signature on the endorsement area on the bank of the check - this tells the bank that you are its customer and gives it an avenue of recourse in the event the check is returned unpaid (i.e. deducting the funds from your account). This is what allows you to negotiate checks that are from different banks at your bank. Not writing your account number on the bank of the check won't prevent the bank from using your account as recourse, but writing it makes things easier on the teller who is cashing the check.
