Well, that depends on what you like in a game.
If you like being a superhero with electric powers, and choosing whether you want to be good or evil, choose inFAMOUS. However, inFAMOUS 2 is now out, so I'd consider that one instead, or you can get both.
If you want a grand, cinematic adventure with stunning visuals and a captivating story, choose Uncharted 2. This is one of my personal favorites, but it depends on what you like.
Both are good, but I'd pick Uncharted 2.

Personally I'd go for Infamous - Assassin's Creed didn't really do it for me. Uncharted 2 is better in terms of physics and graphics than the first Infamous, but having played the demo for Infamous 2 I can say that it's been vastly improved in these terms.
