Until any remains are found, it is most likely that we will never know the fate of either him or his brother, King Edward V. However, there is almost no doubt that they were either murdered in the Tower of London, or died there from unknown causes.
The tower became known as the Bloody Tower in the mid-16th century because it was believed to be the place where the two young Princes in the Tower were murdered by their uncle Richard, Duke of Gloucester.
They were murdered. Most sources say that they were suffocated by feather beds (pillows). However some other sources say that they were drowned in wine OR poisoned with a venomous potion. Richard III betrayed his nephews, Edward V and Richard, Duke of York, as part of his ambition to seize the English throne. After the death of his brother, King Edward IV, Richard, who was the Lord Protector for the young Edward V, took control and declared the boys illegitimate, effectively eliminating them as rivals. This betrayal was driven by Richard's desire for power and the crown, leading to the infamous disappearance of the princes in the Tower of London.
Do you mean why was he responsible for their disappearance? The lost princes disappeared almost immediately after Richard took the throne and it is unknown whether they were killed before or after he left the tower to go on progress following his Coronation. While many theories abound as to who killed the princes, there is no concrete answer. Their bodies have never been verified and no one, to this day, knows what really happened. It is, however, likely that, if he did not order them to be killed himself, Richard probably played a part in their disappearance. Having just taken the throne on rather shaky grounds, and having declared his nephews bastards and therefore not in line for the thrown, he would have wanted to solidify his position as much as possible. Though declaring them bastards had justified him taking the thrown for himself, there would have been nothing to stop the people from rallying behind young Prince Edward as he grew older, particularly if they found Richard to be, in any way, an inferior king. Not only would killing his nephews have ensured Richard's claim to the throne, it would have prevented the people from claiming the princes as their figureheads in later rebellions.
Because Richard III fought a battle with Elizabeth's grandfather, Henry Tudor, in which Richard was killed (the Battle of Bosworth) and Henry took the crown as Henry VII. Richard was supposed to have been responsible for murdering the boy King Edward V and his little brother - the so-called Princes In The Tower. (Young Edward and his brother were the brothers of Elizabeth of York, Henry VII's wife). Therefore the Tudors hated Richard III and would be pleased with any propaganda against him. Shakespeare's play Richard III, helped to give Richard a bad name!
The pretenders to Henry VII's throne were Lambert Simnel and Perkin Warbeck. Lambert Simnel claim was through the support of the Yorkist Dynasty to be young Edward of Warwick ( a nephew of Edward VI and Richard III). Perkin Warbeck's claimed to be Richard of Shrewsbury, Duke of York, the younger son of King Edward IV, one of the Princes of the Tower and was threat to the Tudor Dynasty and gained his support outside of England.

Richard hires people to act out the murders so that he can disassociate himself with villainous behavior.
