Once you catch the 26 letters of the alphabet in Unown, the Maniac Tunnel on Route 214 will be complete, allowing you to catch Unown in '!' and '?' forms. Other than that, nothing else is unlocked.

Catch them and talk to the guy again. This will complete the unown side quest, but that's about it. Nothing really special unless you just really have to have all 28 unown.

Go to the end of the cave where there is another entrance. Go through, and when you get inside, you will see a writing on the wall. It says, "?!" Those two Unown are in there. Capture them. You now have all the Unown.

In Heartgold and Soulsilver, you can now catch the last 2 unown (the "!" and the "?") in the Hall Of The Ruins Of Alph. Then the scientist that gave you the unown recorder comes out and gives you a pat on the back...
