what is going to happen is that your system wont boot again .and if you have bootlock enabled you may restore but if you dont you will have to buy a new bios chip .

But it's highly recommended to unplug your computer while upgrading, cleaning and so on. If you insert the card while your computer is running it can easily fry the motherboard and power supply. All of what was said above is true if we are talking about PCI, -E, -X, and so on. If you are talking about USB, fireware, and so on. Then it's fine.
