To put it simply, it is an acknowledgement of compatible protocols being used for communication. The squeaks, squawks, and bongs of the old analog modems of yesteryear were the handshake process they used to determine carrier speed, the number of data and start/stop bits, parity, and more. Once everything is agreed upon by both ends, the handshake is established and the flow of communication can begin.
