when Jacob shows charlie what he is he said charlie was very shocked and that when he calmed down he expresses interest in the cullens. since then it seems charlie had become very wary of Jacob and shudder's around him sometimes.
Charlie doesn't fully find out what the Cullen's are, he knows they are different from humans but doesn't know and makes his own guesses which the reader's are led to beileve are false. after finding out that he's surrounded by the supernatural such as Jacob and the cullens he expresses to them that he has to know is the absolute mininum and wants none of the details.
its pretty understandable reaction to find out that your best friend's son is a giant wolf so this is why charlie perfers to stay away from him. he isn't like this with the cullens and although he knows they're different he seems to treat them as nicely as he would have before he knew.

Yes, Jacob does know. That is why in the books (I don't know about the movies yet because New Moon isn't in theatres yet) he calls them bloodsuckers and stuff because of a legend that involved his great grandfather finding out the Cullens secret while they were hunting on his land.

In Breaking Dawn, Irina betrays the Cullens by telling the Volturi about Renesmee, Edward and Bella's half-vampire daughter. She does this because in New Moon, Irina's mate, Laurent was going to kill Bella. Jacob and his werewolf pack intervene and kill Laurent first. Irina is still upset about this in Breaking Dawn, so she betrays them.
