Ezio is the ancestor of Desmond Miles (the main character) who accesses Ezio's memories with the Animus. Ezio Auditore da Firenze (Florence) is a Florentine Noble until his father and brothers were killed in the process of a Templar conspiracy. He dons the robes of the Assassin, and hunts down those who had a hand in the execution of his family in a lust for vengeance, ultimately becoming a battle-hardened Assassin. Infiltrating the Vatican and passing up the chance to kill Rodrigo Borgia who had orchestrated everything (Ezio had seen enough bloodshed) and reveals to Desmond through the Animus the message from ancient beings called the First Civilization. In Brotherhood, Ezio becomes a true Master Assassin, liberating Rome from the oppressive rule of the Borgia after his uncle Mario is killed and the Villa Auditore is destroyed in an attack only days after Ezio's assassination attempt of Rodrigo. Ultimately, Ezio establishes the Assassin's Guild in Rome, annihilates the Borgia, and travels east to find out the answers to a letter written by his father, who was also an Assassin before his untimely end. In Revelations, Ezio searches the city of Constantinople, heart of the Ottoman Empire, which has been put under strain by the Templars, who call themselves the Byzantines. Once more, Ezio builds up the Assassin Order with the help of Yusuf Tazim, gathers the 5 mysterious Masyaf Keys with the help of Sofia Sartor to open Altair's library, which reveals the Apple of Eden Altair was in possession of. Ezio finally renounces his life of an Assassin, having spent almost 30 years of his life eliminating Templars. He settles down with Sofia in a Tuscan villa, grew crops to sell, raised two children, and eventually died at "home", in Florence from a heart attack during a visit to the market with Sofia and his daughter.
Strangely, Ezio dies in the same area where his father and brothers were killed during the Pazzi conspiracy.
By characters, you probably mean templars. Templars are the assassins that Desmond sees in Ezios memory. You will get an assassination contract to assassinate the templar. You can also play as several Templars in the multiplayer of Brotherhood.
