The Church of the Holy Sepulcher is located in Jerusalem and is said to be built on top of the Hill of Calvary - where Jesus was crucified. Throughout history, many sects of Christianity have fought over control of the Church and so the keys to the Church have been in the possession of a Muslim family (Nusayba) to keep control neutral.

I have never heard of a Holy Sepulchre of the Blessed Virgin Mary, if someone was using this term, it would probably be an (incorrect) reference to the tomb of the Blessed Virgin Mary which is where her body lay briefly during her dormition before she was assumed into heaven. The Church of the Holy Sepulchre is in Jerusalem, located where Helena, mother of the fourth-century Emperor Constantine, claimed to have located the the very spot where Jesus was crucified. Just a very short distance away, she found the place where Joseph of Arimathea had Jesus buried in his own tomb, although she never explained why a rich and influential man would wish to be buried at a place where the Romans frequently executed criminals.
The church spans the two sites and its construction, intentionally or otherwise, required the destruction of an important temple to Aphrodite. Apart from this, it is very unlikely that the Church of the Holy Sepulchre really marks the place either of Jesus' crucifixion or his burial.
