Take the Uptown 1-2-3 (the red line), 1 stop, from 34th Street-Penn Station to 42nd Street-Times Square.Transfer at 42nd Street-Times Square to the Uptown 7train (the purple line). At Grand Central Station, get the Manhattan-bound 7 train (the purple line), one stop, to 42nd Street-5th Avenue.This station is connected to the 42nd Street-Bryant Park Station (at 42nd Street and 6th Avenue) on the B-D-F-M, the orange line. So you can either exit at 42nd Street and 5th Avenue, and walk one block west above ground, or walk through the tunnel that connects the two stations, and exit at 42nd and 6th.

The Estops at 42nd Street-Port Authority Bus Terminal (at 8th Avenue) but there is a tunnel connecting the 42nd-Port Authority station to the 42nd-Times Square station, so you can transfer without exiting the subway system.
