Though the Sabbath is technically holier in certain ways, Yom Kippur is the usual answer. This is the Day of Atonement, once a year, a full-day fast (Leviticus ch.23) with special prayers and Torah-readings. The Talmud refers to it as The Day (Yoma), or The Great Day (Talmud, Rosh Hashanah 21a); and in Halakha (texts of Jewish law) it's known as the high point of the Days of Awe (Yamim Nora'im). Yom Kippur is the most important holiday because of its special power of atonement.
