Through years and years of research, biological study, and another span of months or years of precise, diligent hard work in a state of the art Bio-Laboratory facility...

Its a deceptively complex question... Many Institutes/facilities even employ different techniques, methods, and procedures, depending on who you ask, or what you are trying to do...

What is the intended purpose/function/outcome of your Transgenic organism? Stating what the organism to be worked with/on is, is crossed/mashed with, or what the combination is meant to accomplish, would make answering this question a large amount easier to answer.

And just 1 warning, with transgenics, as with all genetic experiments, etc... CONTAINMENT is an absolute must. Do whatever you want with your experiments, just keep your work quarantined&contained! Dont let your experiment do whatever IT might want to do... cuz u never kno when ur mutant strain of frost-resistant potatoes might decide its time to take over and dessimate the world agricultural industry, etc. Play SAFE out there. ;)

I myself have brainstormed up some useful, allbeit unorthodox or counter-intuitive crosses of different animals, but have never pursued any of my transgenics ideas, being stuck mostly in basic life-sciences and Biology exploration atm. contact me if u wanna bounce more idea around about transgenics, the applications/dangers of such technologies, etc. Im always up for that kind of discussion. Though it is more complicated than this, scientists basically cut a small section of the DNA out of one organism and insert it into the seeds of an organism of another, unrelated species. For example, inserting a fish gene into a tomato, a bacteria gene into corn, etc.

Getting DNA into an organism is a tough challenge and there are various ways depending on the target organism.

Here are some examples depending on the target:

  1. Bacteria and other monocellular organisms: Certain types of bacteria willingly take up DNA from their surroundings and incorporate them in their genome. These are called "competent cells". Other types of bacteria can be made competent, usually by punching small holes in their cell wall and membrane - for example using electricity or chemicals.
  2. Cultured cells originating from a multicellular organism: In a dish, getting DNA into mammalian or insect cells is similarly simple and usually relies on perforating their cell membrane. Again, electricity or chemicals can be used for this. Success rates are much lower than in bacteria, probably because these cells do not have any machinery intended for taking up foreign DNA.
  3. Plants: A commonly used method relies on Agrobacterium. These bacteria are able to incorporate their DNA into the DNA of a plant they infect. The desired DNA can be introduced into Agrobacterium (see point 1) and then into the plant. In order to generate a transgenic plant, the infected cell needs to be capable of generating an entire plant - e.g. a germ cell.
  4. Animals: There are various ways of generating transgenic animals, which work better or worse depending on the type of animal. For most animal species that are commonly used in genetic research, it is possible to directly inject a liquid containing foreign DNA into a zygote (a fertilised egg) or early stage embryo. Some cells will incorporate the DNA in their genome, with varying rates of success.

In all cases, a selection step is necessary after the treatment because healthy there are always some cells that refuse to take up foreign DNA. Therefore, the introduced DNA needs to contain a selection marker - such as resistance against an antibiotic, or a visible feature of the grown organism (which then has to be manually selected).
