You test the serum of febrile patient for typhoid fever. Then you dilute it to 1 in 80. Means you take 1 ml serurm and 79 ml distrilled water. Accordingly you go on diluting it to 1 in 120, 1 in 160 and 1 in 320. So to be tested till positive in highest dilution. Isolated test positive has no clinical relevance. 'Rising titre' one week apart in febrile illness indicates that the fever is likely to be typhoid fever.
