It depends which route you take. If you're pressed for time and want to do the drive in one day, then the most direct route takes you 10 hours along the Hume Highway and there's really nothing much to see as the highway has been upgraded to dual carriageway and bypasses all but Holbrook at the moment (the Holbrook bypass upgrade has started though, and is expected to be completed by 2013). If you have the time you could vary this route by heading inland at Albury, taking in Yarrawonga, Echuca and Bendigo before heading to Melbourne.
The coastal drive is magnificent, but significantly longer, taking in Kangaroo Valley, Eden and Lakes Entrance. You'll drive this route comfortably in 2 days, breaking your journey at Bega or Tathra, but if you have the time (and the inclination), you could add another day to fully savour the Emerald Coast in all of its picturesque beauty.
