The health officials of Mantua wouldn't let anyone get in or out of the the city because of an infectious plague. This prevented Romeo from hearing about Juliet's plan.
Romeo will know that Juliet is faking her death as part of a plan to be with him, but he is unaware of this plan and believes it to be true.
Friar John was to tell Romeo the plan, but Mantua (the town Romeo was staying in) was in quarantine so he couldn't get to him.
It is Juliet's soliloquy - "O Romeo, Romeo! Wherefore art thou Romeo?" It keeps Romeo there and lets him know that she loves him too.
