Acid-fast staining is due to the high lipid content (mycolic acid) in the cell wall. Cells that do not have these mycolic acids do not absorb the carbolfuchsin. Microorganisms that have taken up the carbolfuchsin are not easily decolorized by the acid-alcohol step in the preparation procedure.
