The contractions of the heart muscle working against the heart valves make the "lub-DUB" sound that we hear as heartbeats. The sounds are made by the closing of :

1) the atrioventricular (AV) valves that separate the atria and ventricles of the heart

The tricuspid valve is closing the entrance from the right atrium to the right ventricle, and the mitral valve is closing the entrance from the left atrium to the left ventricle.

2) the semilunar valves that control the exits from the ventricles

The right ventricle is pumping blood into the pulmonary artery through the pulmonary valve, and the left ventricle is pumping blood into the aorta through the aortic valve.

The two heart sounds therefore mark the entrance (lub) and exit (DUB) of blood from the ventricles.
