After they meet at Brutus's house they plan to murder Caesar the next day.
Whether someone is a villain or a hero in Julius Caesar is a matter of perspective. If you sympathize with Brutus and Cassius's opposition to dictators like Caesar and a return to a more democratic form of government, then they are the heroes and Caesar and Antony are the villains. But if you consider the conspirators to be reactionary terrorists who try to overthrow the government of Rome by the murder of one of its most talented and admirable citizens, then Caesar and Antony are the heroes and Brutus and Cassius are the villains. The play is not committed to either perspective.
The target was Caesar, and Antony was diverted away from the scene when the assassination took place.
Antony just wants the support of Brutus' followers so he can have complete control over Rome.
The extended metaphor Brutus uses to describe how Caesar may act if he is crowned king is the ladder.... Caesar climbing the ladder of prosperity, he would reach the top. He may forget bout the people of Rome. He will look into the clouds and indulge in the wealth and fortune.

I believe that Brutus was a little ticked off in way, because even though Caesar was powerful, he was physically weak, and had many others things gone wrong with him, such as epilepsy for instance. You can see if this is true or not in Act I i believe, if not Act II, but Brutus talks about how he had to get him from the Tiber river, and how Caesar had a seizure, and was shaking on the ground and such, and the foam was coming out of Caesar's mouth and everything, he was screaming for Brutus's help. And Brutus of course went and helped him out.

Getting brutus behind them, claiming to much power
Brutus didn't want Caesar to become king because Rome was a republic. A Republic is a state or country that is not led by a hereditary monarch. Having Caesar as King would ruin Rome as a republic and Brutus didn't want that.
