The best tool for the job would be an incident light meter, to measure the light that falls on the subject rather than the light that reflects from it. Most hand held incident meters are also reflective meters. Failing that, an 18% gray card and a meter reading of the entire card (does not have to be in focus; taking care not to cast a shadow on the card) would also be good.
