I suggest you watch Fullmetal Alchemist Brotherhood first. It is better to understand the basics of the story first than to suddenly jump on the movie. Otherwise you will get confused by the many characters and the terms in the movie. But, if you have watched Fullmetal Alchemist (the first Fullmetal Alchemist anime, it has a big difference with Brotherhood but the basics of the story are the same) you should just go for the movie, since you are already familiar with the characters and terms.
You should see the last three episodes of fullmetal Alchemist at least to understand certain things mentioned in the movie. Actually you should watch the entire series to understand the conflict and what's going on. My advice to you just watch the series first. :)
If you like quite happy endings better than a quite sad one I suggest you watch brotherhood first because in the end winry and ed are a happy family. Even though ed has lost his ability to use alchemy.
The other full metal alchemists has a rather sad ending in the conquerer of shamballa which ends with ed and al trapped in our world forever!

But there is a movie continuation to Fullmetal Alchemist, called "The Conqueror of Shamballa", which concludes the story. There is also another movie that supposedly takes place during Fullmetal Alchemist; Brotherhood episodes 20 and 21 called "The Sacred Star of Milos". However, it doesn't fall into place very well with the story.
