The most significant difference is just historical. Shia believe that the first Caliph entitled to rule after death of prophet Muhammad was his cousin Ali Ibn Abou Taleb and the husband of the prophet daughter. However, Muslims elected Abu Bakr, the prophet companion during his Hijra trip to Medina and the first man who converted to Islam, to be the first caliph. No other crucial differences.
