You must take Amtrak to Back Bay Station in Boston. Then you can either walk, take a cab, or take the Orange Line subway to the Mass Ave station and walk from there.
It's 4 stops on the Uptown E train, so it's about 5 to 10 minutes.
Take the Downtown A-C-E (the blue line) from 42nd Street-Port Authority to West 4th Street.Transfer at West 4th Street to the Downtown F train (of the B-D-F-M, the orange line) to 2nd Avenue-Houston Street. Then walk 4 blocks up 2nd Avenue to 4th Street. After Houston comes 1st, 2nd, 3rd, etc.
Get on the Southbound subway from Yorkdale Station (which is connected to Yorkdale Mall) and get off at Spadina subway station. From there take the 510 Streetcar and get off at Sussex Ave or whichever is your stop. You can also get to U of T from St. George Station, Queen's Park station or Museum, but I am not sure which bus you would take from those stations. !

Take the Uptown 2 or 3 express trains (of the 1-2-3, the red line), 2 stops, from 34th Street-Penn Station to 72nd Street (at Broadway/Amsterdam). Broadway and Amsterdam intersect at this point.
