Lady Macbeth advised Macbeth not to say "amen" after committing the murder because it would make their deed more real and harder to go through with. She believed it would be better to keep their guilt hidden by not acknowledging it through prayer.

Macbeth killed King Duncan and the two royal chamberlains. He then advised his wife of the murders. He was unhinged by the bloodiness of the killings, and by his inability to say 'Amen' to the prayers of the dying chamberlains.
In Act 2 Scene 2, Macbeth [c. 1014-August 15, 1057] killed King Duncan I [d. August 14, 1040] and his two royal guards. He reported the deed to his fellow conspirator and wife, Lady Macbeth [b. c. 1015]. He was unhinged by the bloodied corpses and the bloodied crime scene. Particularly was he troubled by his inability to say 'amen' to the guards' prayers before being stabbed to death. Macbeth's Lady told him to stop thinking or go crazy [Lines 13-14].
The word that stuck in Macbeth's throat as he stood over the servants was "amen." He couldn't say it because it represented a prayer for their eternal rest, which he had denied them by ordering their murder.
At this point int the play, Macbeth has just committed a terrible crime: murder. Back in those days, Catholicism was big, and "Amen" was like a nice little blessing to finish off a prayer. We know that Macbeth eventually goes mad over what he has done, and this is where it may have begun. He is so ashamed of the terrible thing he has done, that he believes himself to be undeserving of the blessing of the word "Amen". Murder, being a disgrace to God, would have been almost unforgivable in those days. I also absoloutly freakin love chocolate icing on a chocolate caramel and custard cake. Like sereals WOW!
Christianity does not make an appearance in Macbeth. The closest it comes is when Macbeth reports that the grooms woke up, said prayers and went back to sleep, but Macbeth could not say "Amen". This symbolizes that his guilt is standing in the way of his salvation.
Macbeth was clearly disturbed by the murder of Duncan and the fact that he was unable to pray (to pronounce the word "Amen") when the grooms prayed. This caused him to forget the plan that Lady Macbeth had concocted.
