In Twilight, Jacob is unaware that the Cullen family are vampires. He just knows that the Cullen's and the Quiluete tribe do not get along and that the Cullen's are not allowed on the reservation. He believes that the "vampires of forks" are just a scary story told by the elders in his village.
It isn't until New Moon, after Jacob undergoes the transformation into becoming a werewolf, that his father tells him the truth about the Cullen family. It is because of the Cullen's and the other vampires, that the wolf gene is triggered anyway.
