Which .45 handgun are you referring to? The .45 Colt revolver (Peacemaker) goes back to about 1873. The traditional "45 Auto" was adopted by the US Military in 1911, but there were other, earlier .45 caliber automatics. And of course, .45 caliber black powder ball and cap pistols, ball and cap revolvers, etc are MUCH earlier than the Peacemaker. So... which handgun?

On the barrel of most guns produced, the proper caliber/gauge is stamped as well as size/ type. That being said I think the model 16 was a semi-auto .22 caliber, firing a .22 caliber long rifle round.
Generally it is a small concealable auto loading pistol of limited power. It takes its name from the .25 ACP cartridge that it fires. It is called .25 caliber (notice the decimal point?) since the bullet it fires is about 25/100ths of an inch in diameter.
