Technically at the University of Colorado, I believe, because Matt and Trey were taking a film class there and they had to make a short. So they created, "The Spirit of Christmas" in it there features two characters who are very similar to Stan and Kyle and two other characters that look like Kenny and Cartman except that Cartman is named "Kenny" and he therefore dies and the whole "Oh my god they killed Kenny! You bastards!" thing was said to him instead of the Kenny we know and love now.

In 1996 they then went on to create another christmas-based short titled 'Santa vs. Jesus'. They finally began Season 1 of South Park in 1997, I hope this answers your question.
