In Pokémon Black 2, in order to be able to go to the Nature Preserve, you will first need the accomplishment of seeing all Pokémon within the Unova regional Poké Dex. Once you have accomplished seeing all regional Pokémon then you can go to Professor Juniper's Lab in Nuvema Town in order to talk to her and she will give you the Permit which will allow you access to the Nature Preserve.
You need to recieve the Permit first before you can fly to the Nature Reserve from Mistralton City. You must have seen all Unova Pokemon in order for Professor Juniper to give you it.
