Jesus brothers were James, Joseph Jr., Simon, and Judas. No John. (reference Matthew 13:55 and Mark 6:3.)

If you're talking about John the Apostle, in John 19, when Jesus was on the cross, "he saw his mother there, and the disciple whom he loved (John) standing nearby, he said to his mother, 'Dear woman, here is your son,' and to the disciple, 'Here is your mother.' From that time on, this disciple took her into his home."He was not saying that they were really mother and son. He was telling John to take his place in taking care of his mother.

The bible clearly says that Jesus mediates for us in 1 Tim 2:5; Heb 9:15 and Heb 12:24. The Bible also calls Jesus our advocate in 1 John 2:1.
