On ultimate ninja, you dont, you get to play as him on ultimate ninja 3, and you unlock him by unlocking everyone else first. a shortcut for this is to put in the every character ulocked cheat, then beat the rpg mode, then unlock hanabi, then it will say the yellow flash is now available in Vs. Duel

Well, I can't find Sasuke myself (ran through the whole map, except the Hokage's roof top) but I found the Third Hokage in the Training Fields, after I completed Shizune's request. The medicines can be found easily, if you use Naruto's Shadow Clone ability or Neji's Byakugan. Classic sasuke is in the first room at the right in orochimaru's hideout. ( you can't get him till after you finish the sasuke RPG). The shipudden Sasuke is in the first room of the Chambers of Darkness ( after you go through it with both Sasuke , and Naruto in the Chambers of Darkness the black chronicals and the Chamber of Darkness the White chronicals)
