Staphylococcus aureus is negative for motility, and positive for the indole production. The SIM test tests for sulfur reduction, indole production and motility. Positive for indole production means tryptophan is broken down into indole and pyruvate and will give a red color. Motility, you will see the bacteria move outside of the stab. Why is the production of an isolation streak key to the identifcation of a bacterium grown in culture?

Isolation streaking was first introduced by Robert Koch as an improvement on the dilution method of isolation of bacterial cultures. Isolation is achieved by taking a minute sample from within one bacterial colony and putting it in a sterile medium so that only that organism will grow.
