The S/PDIF port is used to connect the graphic card to the motherboard in order to get sound in some systems.

Each sound card can be different, but usally it can be the blue port on the soundcard. If you dont have a blue port try to idenity the "line in" if you are using any TV turner software you will have to configure the software to get the sound from "line in" in the configuration. The blue port is most likely used for a microphone line-in.
