It cost three cents well into the 1940s. In the late 50s, it rose to four cents... and these days, it's not quite fifty cents-- it's currently forty-nine cents. But if you are asking when the first fifty cent definitive stamp came out, that was probably in early November 1894: The US Postal service says this stamp appeared in the first definitive series printed by the Bureau of Engraving and Printing, and it showed an image of Thomas Jefferson.
