The lead actor of The Lord Chamberlain's Men, who had exclusive rights to perform Shakespeare's plays after 1594, was Richard Burbage. Burbage was recognized as the second greatest actor of his day, after Ned Alleyn of The Lord Admiral's Men. Burbage was not only the lead actor but also the main shareholder in the company and the largest shareholder (along with his brother Cuthbert) in the Globe and Blackfriars theatres.
An anonymous contemporary poet wrote that Burbage played Hamlet, Lear and Othello. A contemporary joke is based on the fact that he played Richard III.
"Best" is of course a subjective thing, but the most popular are Richard II, the two parts of Henry IV, Henry V and Richard III.
This is a matter of opinion, but Hamlet and King Lear are usually the plays mentioned as Shakespeare's best.

Since we are not exactly sure of the dates of composition of any of Shakespeare's plays, this is a bit of an estimate. However, the years 1594 to 1610 could be roughly considered Shakespeare's best.

The one that he acted best in, of course.
Do you mean which play was first or which play was the best of all the Shakespeare plays which were performed at the Globe? If the latter, it is really just a matter of opinion. If the former, a lot of people think the first play to play at the Globe was Julius Caesar.
