There is no character of Queen Elizabeth in the play Romeo and Juliet. You might be thinking of the movie Shakespeare in Love, which is about the writing of Romeo and Juliet, and in which Queen Elizabeth is indeed a character in that film.

Many people only think of this question superficially. Although the Queen is never mentioned, her influence is definitely there. She ruled with strict morals, and Shakespeare's characters directly violated them. The fact that characters are self-destructive may mean that Shakespeare was describing what can happen when people go against these morals.
