He absolutely can! The question becomes, will he. The most likely scenario in which he would run in 2012 is if the next President is a Republican. If the next President of the United States is a Democrat, he still could, but most likely would not run against the Democrat incumbent in 2012. Under this scenario, he would be much more likely to run in 2016. While this may seem like a long time from now, it is important to note that he would be 63 years old. By contrast, Barack Obama is 46 years old, Hillary Clinton is 60 and John McCain is 71. In having to wait 8 years, it is also likely that Edwards would be able to gain political capital and experience by serving as a Vice President or Cabinet Level Position.
As of now, men are most likely to win, but any gender is allowed to run.

No, Obama can't run for president in 2016. This is his second term and the constitution clearly states that a president may only be in office for a maximum of two terms. Obama was never going to run for president in 2016 whether he won to Mitt Romney or not. Joe Biden and Hilary Clinton will most likely be the Democrats running in 2016.
