It is possible but highly unlikely. She is 10th in line to the throne, and will be pushed back further if any of her siblings or their offspring have any more children. Something would have to happen to make all of those unable to take the throne.
(There have been some changes proposed and agreed on, to change the rules of succession. One is to end male preference primogeniture, which meant a younger son would supplant an older daughter on the line of succession. However even when the change is fully enacted, it will not be retroactive, Anne will not move ahead of her three younger brothers.)
It depends. As of now she is not. Because Charles is Prince of Wales if he outlives his mother Camilla will be the next Queen, but if Charles dies before his mother Catherine will be the next Queen.

Princess Anne, now known as the Princess Royal, and Prince Andrew, the Duke of York, had lower media profiles because they are not direct heirs to the throne. Andrew is fourth in line, while Anne is 10th. It was the same for their younger brother Prince Edward, the Earl of Wessex, who is seventh in line. Much of the media attention at the Diamond Jubilee events went to the family of the Queen's eldest son, Prince Charles, who is the direct heir to the throne.
Prince Charles is 2 years older than Princess Anne.

No King has ever ended royalty. During the 20th century there have been some changes as to titles, but they never got rid of royalty. After WWII in England to cut down on the number of Prince and Princesses they decided that only children of males directly in line to the throne can be Prince and Princesses. This is why Prince Andrews children are Princess while Princess Anne's children do not have any titles. The one exeception to this was when the current Queen Elizabeth gave birth to Charles and Anne. Because the way the rules were set up Charles and Anne would have been styled Lord Charles and Lady Anne Montbatten-Windsor, but because Elizabeth and Charles were and are the heirs to the throne Charles and Anne are Prince Charles and Princess Anne.
