I am Not sure how exactly you define "prayer". If you are simply saying the first time Man talked to God then that would be Adam. Although he definitely spoke to God before this, the first time it is recorded that he talked to God was in Genesis 3:9
There are many times from Genesis 3 and on where man talks to God, but if you mean "prayer" as in asking God for something then that would be in Genesis 15:8 where Abram asks God for a sign that all the things that God told him would come to pass.
