Tybalt wants to fight him. Tybalt always wants to fight someone, and he has fastened on Romeo after Tybalt spotted him at the party in Act I Scene 5.
Mercutio starts the fight that ends with Romeo fleeing the city.

His name is Mercutio. Mercutio is a friend of Romeo's and a relative of the Prince's. He does not exactly take Romeo's place in a fight: Tybalt wants to have a fight with Romeo but Romeo refuses to fight him. Mercutio, who has been taunting Tybalt right along, thinks it a shame that there is to be no fight and is disappointed in Romeo for refusing to take part, so he challenges Tybalt himself.
Romeo picks a fight with Tybalt in Shakespeare's play "Romeo and Juliet" after Tybalt insults him and his friends at a party. Tybalt challenges Romeo to a duel, but Romeo refuses to fight. This leads to Mercutio, Romeo's friend, stepping in and fighting Tybalt instead, resulting in tragic consequences.
"Give me my rapier, boy." It was a chance for a fight.He is extremely rude and wants to fight him. He assumes that somehow Romeo's attendance at the party is intended to insult the Capulets. For Tybalt, anything will do for an excuse to fight.
The first time he hears that Tybalt wants to fight him in a duel, Tybalt is his cousin by marriage, although only Romeo knows this.
Tybalt turns on him and begins hitting him, Romeo does nothing because he says he loves Juliet and so he must love Tybalt (her cousin) too. Mercutio steps in to keep Tybalt from killing Romeo but in turn is killed by Tybalt.

In this scene Tybalt seeks Romeo because he is mad at Romeo for being at the party at the Capulet house, and wants revenge. In the play, this is the only reason but in the movie, Tybalt is mad at Romeo because he knows Juliet is falling for him.
