Friar Lawrence sends a message by Friar John to Romeo in exile in Mantua because the whole exercise of drugging Juliet and getting her buried in the tomb would be pointless if Romeo wasn't in on the plan so he could come and scoop her off to Mantua.
Well, we don't know exactly what it said, but its gist was that Juliet was pretending to be dead and that Romeo was to show up at the Capulet tomb at a certain time, when Juliet would wake up and he could take her to Mantua.
The letter explains that Juliet intends to take a drug to induce a coma, faking her death, so that she and Romeo could run off together without anyone looking for her.
Juliet sends the Nurse and her servant Peter to Romeo to learn his arrangements for the wedding.
None, unfortunately. Romeo was waiting for news from the Friar, and the Friar had sent it, but it got lost in the mail (specifically, the courier got locked up in quarantine).
He sends a message to Romeo by way of his fellow friar, Friar John.

Mantua is where Romeo is. He's been banished there for killing Tybalt. The friar has to send a message to Romeo so that Romeo will find out about the plan for Juliet to fake her death, or Romeo will think Juliet is actually dead. ***SPOILER*** The reason Romeo and Juliet died was because the letter never actually got to Romeo.
Somewhere
Juliet sent the nurse to see Romeo about the wedding plans at 9 o'clock in the morning.
Obviously there was no such thing as e-mails when Shakespeare was writing. What you have been asked to do is to imagine what Romeo and Juliet might say to each other if they had the technology to send e-mails. You have to use your imagination to put yourself in their place of Romeo and Juliet. If you haven't got an imagination you are in deep trouble.
