The Capulet and Montegue feud for short. There may been some book evidence that it was over romeo spicifaclly, but over all it was the two families' fued. Except, of course, that Tybalt was not a Capulet (although he was related to one) and Mercutio was not even related to the Montagues.
No, Tybalt had taken it upon himself to be the champion of the Capulets (even though the head of the family told him to knock it off). He decided to take offence at Romeo and had challenged him to a duel. But Romeo did not want to fight him, because Tybalt was related to Juliet. Mercutio then stepped in and challenged Tybalt to a duel, which Tybalt accepted. There was no reason for these two to be fighting except that they were quarrelsome.
For two reasons. First, it physically separates Romeo and Juliet, since Romeo has to go to Mantua and Juliet is still locked up in her parents' house. Second, it makes any hope that the Capulets can accept Romeo as a son-in-law if the marriage were presented to them as a fait accompli extremely unlikely.
Tybalt's death is the turning point because it leads to many dramatic things in the play.
