Our mechanic had to climb through the inside of the car, into the trunk area and open it by removing the hatch cover. That's the only way he could replace the latch.
There is a button you have to push under the steering wheel column. This button is located on the bottom trim about 4-6 inches from the hood release.
Removed the back panel from inside staring from top to bottom of the plastic, and you will be able to open. Then replace the back door handle.
If the handle is broken, then you'll have to manually open it. If the locking mechanism is frozen, you will have to manually open it. Generally that means pulling the back panel off and pulling the latch cables by hand. Sequoia forums have detailed information on how to remove the rear panel - fairly simple.
If you mean the mpx door relay it is in the rear door itself. Open the hatch and remove the plastic cover on the inside of the door. The mpx door relay is a black box with wiring running to it.

open the rear passenger side door, pop up the seat as if you were going to enter the back seat. look for a trap door in the carpet under the seat you just lifted up. open the door, there's the jack

The back hatch will often not open if the internal mechanism or release has become damaged. Replacing the latch is the most effective and reliable fix.
the rod for the door handle is held on with a plastic clip it may have wore out
If it's the gas tank hatch. it could just be the small metal that pushes the cover out once the lock has been released. you can bend this a little more wider. enough to have tension to push the gas tank cover out. It happened on my Sequoia once.
